What your talking about is called depth of field and is more a funtion of the aperature of the lens than of the focus. Typically with a macro lens you have a very small depth of field, meaning only a small area is in focus. No matter how much you focus you will not change the depth of field. All you will be doing is changing the specific area of the bud that is in focus.

The best way to change your depth of field is to change the aperature of the lens. The higher the aperature the greater the depth of field. When I take macro bud shots I use the highest setting on my lens which I think is F32. If you have the ability to change the aperature on your camera this will make a big difference.

Keeping the camera absolutely still will help with the focus a bit. It won't really do anything for the depth of field but when shooting macro anything that can improve the focus is worthwhile. Use a tripod if you have one.
