Those brainscans showing 'holes' in the brain were not holes at all, they were areas of the brain depleted of the neurotransmitter serotonin. Ecstasy works in part by flooding the brain with this neurotransmitter, after a night of rolling your brain wont have much left but this is not permanent and the brain replenishes itself within a week.

Dr. George Ricaurte and his research team at John Hopkins university attempted to prove the neurotoxicity of MDMA by giving it to primates, he concluded that "humans who use repeated doses of MDMA over several hours are at high risk for incurring severe brain dopaminergic neural injury (along with significant serotonergic neurotoxicity)."
He later retracted his paper, because the experiments did not use MDMA but instead methamphetamine.
