This may seem kind of redundant, but when I defrost it should I just take it out the morning of and let it defrost on its own or should I put it in the fridge the night before?

That would depend on how much time you have until your scheduled U/A.

Can I microwave it just before going or just use the hand warmer to keep it from going bad.

Human urine can be microwaved, but I generally dont recommend it because human urine is bio-hazardous and should not be in an area where food/utensils are involved.
