the 6 lines correspond to each string and the number is the fret the note is played on. Whatever tuning you are in the open note of each string is the begining of each notation progression. So lets say you are in standard tuning (e,a,d,g,b,e) and the tab looks like this:

you would play the open note which is an e then the first fret which is an a, second fret a#, and the third fret is a b. Your note is a b. Just know what tuning you are in so you know where to start than just go through the notation progression on up the neck until you find your note.
ultimate-guitar has some program called power tab, where it lets you play with the song. i havent tried it out, but my friend said its really helpful.
Powertab: I'm Pretty sure the new version can convert to Staff.
Guitar Pro: If you can get do so.

I've found a couple of programs out there that let me enter the tab, and it outputs it as a midi file. I haven't tried them properly yet, but I'm sure I can get the result I want from one of them.
