If that collector was Nazi enough to suspect that a sub is being used, she wouldve, under DHHS regulations, ordered you to submit a second specimen immediately, this time under direct observation (with an observer of same gender as the donor). She would then process and send off

No need to worry there. Often, the site of specimen collection and the site of analysis are at two different locations. Not all collection facilities are licensed to perform analysis. Thats why the specimens are sent off. The lab will perform an Immunoassay screen first along with the colorimetric validity screen for dilution/adulteration/substitution.
