Just took a pre employment test and I was going to use the method on the dillution thread so I followed it to a T and after takeing ten times the daily recomended dose of b12 my pee was still as clear as a mountian stream. I went with plan B and used methods described in substitution thread and seemed to have worked she accepted the pee so I will have to wait and see how it turned out. I was curious why they take a sample to look at under a michroscope??? And why wouldnt my pee turn yellow after taking so much vitamin B12?? Thanks for any info or help

Because youve used B12.
The proper vitamin for urine color is B2 (Riboflavin).
I was curious why they take a sample to look at under a michroscope??? The sample will not "go under a microscope". Drug testing is not CSI.
Even though the sample was sent off to a lab for analysis, the lab will still perform an initial Immunoassay for drug metabolites and a colorimetric assay for the validity check (creatinine, pH, specific gravity, excessive oxidants, excessive nitrites, excessive chromates, and the presence of glutaraldehyde).
The site for specimen collection and the site for analysis often occur at two different locations, mainly because the collection facility was not certified to perform analysis.

So with these tests being performed will the pee I used work? It was frozen for about 3 weeks then I thawed it, will this be a problem?

As long as the sub donor was known to be drug-free (that includes scripted/OTC meds), and had not consumed a large volume of fluids prior to giving you the sample, you have nothing to worry about.
And as long as you didnt spike your sample with adulterants containing nitrites, chromium, or glutaraldehyde, then you have nothing to worry about. The use of other forms of adulterants will cause an abnormal pH.
Frozen urine stays "good" for up to 12 months.

The reason I asked about the michroscope was because before she sealed the containers I saw her take some out and put it under a michroscope, she never looked at it while I was there but Im sure she was going to?

Did she seal the containers and bagged/tagged them in your view? (as mandated by DHHS regulations, as well as DOT regulations)
she never looked at it while I was there but Im sure she was going to? What is it that youre worried about?
