yes it will work

its called deep water culture as you seem already to be aware...do a little search on it, i started doing it about a year ago(in buckets, not one big tank like you) and i will never go back...i just love it

if you have one big reservoir, it will not be hard to maintain at all...the only thing i can say initially is depending how big your tank is (cant tell from the diagram) you may need a few more airstones...one under each plant perhaps?

also you will need to black out the aquarium somehow, the roots/tank should not be getting any light

I think you'd be better off with a plastic reservoir. A glass aquarium isn't the best thing going, it's heavy you have to lightproof it and worry about breaking it. Do a search here and you will see lots of people use rubbermaid tubs. I have some that I bought from Home depot, they are 18 gallon ones made from recycled materials, they're black and they come with a lid for under $5. Very nice

The hardest part is changing water and that doesn't have to be. Just depends how you do it. Oh and get the air pump above water level or a backflow device. Just in case the power goes out so it won't siphon back through the pump. That can be messy.

You will also want to make sure there is an airstone under each plant or your roots will rot off. Also get a very powerful air pump. Providing enough air for the roots to survive is tough (at least in my experience) but possible.
